SUMMARY:

Helicobacter pylori (H. pylori) is a bacterium that can reside in the human stomach and lead to the development of gastric ulcers, gastritis and even gastric cancer. Forits treatment it is recommended the use medications such as proton pump inhibitors, a combination of two or more antibiotics (such as metronidazole and amoxicillin) and adjuvants such as bismuth (III) salts. Bismuth is capable of reducing viral replication by inhibiting several enzymes and its therapeutic combination with N-acetyl-cysteine (NAC), forming the Bi(NAC)3 complex, favors not only its stabilization in gastric conditions but also improves its absorption into tissues when administered orally. To eradicate H. pylori, problems related to oral administration, bacterial resistance and drug absorption need to be overcome and for this, the encapsulation of drugs in polymeric systems, especially the nanostructured ones, may be one of the alternatives. Another option would be the increase of the retention time of drugs in the stomach by using gastroretentive formulations, including those with mucoadhesive characteristics. In this way, it is expected to prolong the release time of medications and reduce the frequency of daily administration. The aim of this work is to encapsulate metronidazole and Bi(NAC)3 in a polymer that offers gastro-resistant and mucoadhesive characteristics for controlled and concomitant release of the drugs. For this purpose, encapsulation will be performed with different combinations of polymers and the study of their physicochemical properties, as well as the release profile in gastric pH and the evaluation of in vitro toxicity.
